:D I have that exact photo from when we went there! We planned to spend a long weekend there, but--lovely as it is--we ended up travelling to Montevideo on the second day. Depending on what you are looking for, I would say that a day and a night is a good amount of time to enjoy Colonia.

To echo the above, I have just started reading Open Veins and would whole heartedly recommend it. There's a lot of history and detailed information there, but it is easy to read due to the author's ability as a story teller.
