I have taken the bus to Rio. However it was via a stop in Igauzu. From Bs As we went to Iguazu with Via Bariloche which was brilliant. Full lie back beds, movies, 3 course meal, nightcap, slept very well. From Iguazu to Rio we went with Crucero del Norte. They don't have flat beds, only semi cama. The bus was comfortable, but a long way to go without a bed.
All toursists on the bus i'd say so no problems what so ever. They stop every now and then at these big road houses where you pay for food by weight (very common in Brazil I found!).
I didn't have any concerns for safety... not sure if 40 hours straight would be too comfortable. I'd look at GOL if I went again.
