Since the sign is like 100 years old maybe and written in Spanish from Spain (no?), it could be for sanitary reasons (Churches being the place where people gathered the most at the time):

It's certainly Spanish usage, so perhaps the church had a Spanish priest at the time. It's still to get a sign with similar lettering - we had one made in Buenos Aires a few years ago.

HAHAHAAAAAAAAAAHA i still remember the old tin signs at the train stations telling where to spit!!
 
But religion apart, people here spit anywhere and any time, I used to work in a call centre and Argie beside me used to spit on the carpet and then he used to clean it with his shoes like Cat always cover it's shit... and I have seen people spit in Kitchen sink, I used to share an apartment with an Argie and he used to spit in the kitchen sink always. Even in the bus you can read do not spit, It's like here people made up there mind that if it is not written do not spit, It means it is allowed to spit.
