Does anyone know of a decent house to rent in Uruguay for 6 people over a long weekend. A place they have been to that is easily accessible and close to shops and nature ?

I think that's pretty tough to do. The only temp-rental market I'm aware of is in Punta del Este, which is close to shops, if not to much nature. Most rentals there are apartments by the week or, more usual, 15 days.
A place that might suit your needs is http://www.posada-gondwana.com. It's a small estancia about 20 minutes' drive outside Colonia, way out in the country. In addition to a few rooms in the main house, there's a guesthouse with 3 or 4 very comfortable rooms sharing a big porch. If you occupy all of them, it would be almost like having your own house. The setting is very laid-back but quite comfortable, and the owners are charming. Plus there's a great pool, and they treat you to a terrific breakfast.
